Physiological Characteristics of Histamine Receptor on Thoracic Aorta Smooth Muscle Motility in Guinea-pig

Abstract
|
|
|
To elucidate the physiological characteristics of histamine receptor the isolated thoracic aorta in guinea-pig, effects of H1, H2-receptor blockers and Ca++-Channel blockers on the contractile responses of histamine were investigated in the isolated thoracic aorta of guinea-pig.
The results were summarized as follows
1. Histamine caused the contraction in the isolated thoracic aorta of guinea-pig, and the contractile responses increased between the concentration of 3 x 10-5 M and 2 x 10-4 M in dose-dependent manner.
2. The contractile response induced by histamine was completely inhibited by the pretreatment with Hi-receptor antagonist, pyrilamine, but was increased by the pretreatment with H2-receptor antagonist, cimetidine.
3. The contracility induced by histamine (3 x 10-5 M) was decreased in the Ca++ free medium.
4. The contractile response induced by histamine (3 x10-5 M) was markedly inhibited by pretreatment with papaverine (5 x 10-5 M) or verapamil (5 x 10-5 M).
